Give Burlington one full day, two if you want the Royal Botanical Gardens and a lakefront afternoon. It is a stop between Toronto and Niagara, not a base.
Burlington sits on the north shore of Lake Ontario between Toronto and Hamilton, about 45 minutes by GO train from Union Station and roughly an hour from Niagara. The draw is the lakefront — Spencer Smith Park, the Brant Street Pier and the Waterfront Trail — plus the Royal Botanical Gardens, Canada's largest. Downtown is compact and walkable; the rest of the city is spread out and easier by car. Most travellers pass through for a day or use it as a quieter, cheaper bed than downtown Toronto.

Downtown is walkable and the lakefront trail is paved. For anything beyond that you want a car: arterial roads sit about 2 km apart and the city sprawls. Burlington Transit buses run roughly every 30 min; routes 3 and 5 link Burlington GO Station to downtown, and route 1/1X runs from Burlington GO past the Royal Botanical Gardens toward Hamilton. Cash fare is C$3.50; a Presto card or contactless credit/debit tap costs less, and a GO-to-Burlington-Transit transfer is free when you tap both. GO trains connect Burlington GO and Appleby GO to Toronto; VIA Rail and Amtrak's Maple Leaf stop at Aldershot.
