Skip unless you are here for Semana Santa (Holy Week) or as a ferry hub to Adonara and Lembata. One night is enough.
Larantuka is the Catholic heart of Flores, famous for its Holy Week processions. Outside Easter it is a quiet port town with a cathedral, a few warungs and not much else. Most travellers pass through on the way to the islands east of Flores.
