Santa Cruz
One night is enough for the Colchagua Museum and a wine tour; use it as a day trip from Santiago (2 h by bus) or a stop between Santiago and the south.
Santa Cruz is the gateway to Chile's Colchagua Valley, the country's premier wine region known for carménère. The town itself is small, but the Colchagua Museum is a world-class history collection, and vineyards like Clos Apalta and Montes are a short drive away. Most travellers visit on a day trip from Santiago or stay one night to do two wineries.
