BWI is the practical gateway for most visitors. From the terminal, follow signs to the rental car centre, pick up a car and take I-97 south to US-50 east across the Chesapeake Bay Bridge. The bridge toll is charged eastbound only; check the current rate. From the bridge it is a straight run to Cambridge, with exits for the historic district and the waterfront. Allow extra time on summer Fridays and Sunday afternoons, when beach traffic backs up at the bridge.
Salisbury-Ocean City Regional Airport is much closer and works well if you can find a flight that suits. It is a small airport with limited carriers, so check schedules before you commit. From Salisbury, US-50 west brings you into Cambridge in under an hour. Easton Airport is smaller still and mostly serves private aircraft; do not plan a commercial arrival there.
There is no train station in Cambridge. Amtrak's Northeast Corridor runs through Baltimore and Washington, not the Eastern Shore, so rail plus a taxi is not a realistic plan unless you are already in Baltimore and happy to pay for the drive. Intercity buses do not serve Cambridge either.
